چکیده

The majority of modern organisms, including many prokaryotes, are aerobes;1 that is, they use molecular oxygen as the terminal electron acceptor for energy generation. Although nearly every redox gradient in nature appears to be utilized by one organism or another,2-4 aerobic metabolism predominates, in large part due to the highly exergonic nature of the four-electron, four-proton (4e/4H+) reduction of O2 to H2O (reaction 1). A multicellular aerobe requires an

Spectroscopic determination of cytochrome c oxidase content in tissues containing myoglobin or hemoglobin.

A simple spectroscopic method for determining the cytochrome c oxidase, cytochrome a, a3, content in tissue and mitochondria samples independent of myoglobin or blood contamination is described. Using tissue homogenates solubilized in Triton X-100, this assay relies on the selective reduction of mitochondrial cytochromes by the action of potassium cyanide. Monitoring the optical absorbance of t...

Ligand Binding to Heme Proteins

The 02, CO, and alkyl isocyanide-binding properties of a variety of vertebrate and invertebrate heme proteins have been compared in detail to those of protoheme mOn0-3-( 1-imidazoy1)-propylamide monomethyl ester in aqueous suspensions of soap micelles. The proteins examined include: cytochrome P-450,,, from Pseudomonasputida, beef heart cytochrome c oxidase, yeast cytochrome c peroxidase, a and...

Myoglobin-enhanced oxygen delivery to isolated cardiac mitochondria.

The heart, red skeletal muscles and the nitrogen-fixing legume root nodule function in steady states of high oxygen influx, partial oxygenation of cytoplasmic myoglobin or leghemoglobin and correspondingly low oxygen partial pressure. Here, we ask: what conditions are required at the surface of actively respiring, state III, tightly coupled mitochondria to enhance oxygen flow to cytochrome oxid...

Myoglobin and Mitochondria: How Does The “Oxygen Store” Work?

The most important condition for functioning of all living aerobic organisms is continuous supply of sufficient oxygen. The access of oxygen to metabolic processes in cells is mediated by the respiratory proteins, tetrameric blood hemoglobin (Hb) and monomeric muscle myoglobin (Mb). Myoglobin is expressed in red muscles in response to mitochondrial demand for oxygen, transporting it from sarcol...

A theoretical study on the binding of O(2), NO and CO to heme proteins.

The hybrid density functional B3LYP is used to describe the bonding of the diatomic molecules O(2), NO and CO to ferrous heme. Three different models are used, a five-coordinated porphyrin in benzene, the myoglobin active site including the distal histidine and the binuclear center in cytochrome oxidase. The geometric and electronic structures are well described by the B3LYP functional, while e...
